Valve is taking a break from anything and won't be at E3.
I make E3 topics for all the Keynote conferences every year.
But I'm looking forward to E3. It's going to be fun.
Nintendo are probably going to steal the show with Project Cafe, but that makes me all the more interested in what Sony and MS have to show off. Seeing as they know Nintedo are bringing out a new console, they've got to show something off to not be one-upped.

Welp, I'm excited. E3 starts tomorrow, but we have to get the boring conferences out of the way first (lawlz)!
It's not every year that you see an entirely new generation. Maybe stupid sites will finally stop referring to the current generation as 'next-gen'.

Speaking of which, I need to make an E3 predictions post, anyone want to start it off here?
I'm calling (wishing for) a new gorgeous F-Zero for Cafe, Pikmin 3 on Cafe (duh), Skyward Sword stays on Wii, Four Swords 2 as a launch title for the next download service, and whatever Retro Studios shows off will officially be the best thing ever.
Really hope but probably won't happen: Okami 2 for Cafe melting everyone's eyeballs with beautiful amazingness, a new Star Fox using Cafe's controller screen as the communicator screen, and The World Ends With You 2 for 3DS (I would do many illegal things to make that happen). L.A. Noire with much better facial meshes so the MotionScan tech doesn't lose all the subtle facial movements would be nice too.

@Brad: Somehow I feel SS will end up like TP on the GC. They'll hold it back for the release of the new console or something.
But there are rumours abound of a 3D Pokemon (possibly MMO too)... DO WANT. A 3D console Pokemon game is a long time coming - I'd prefer if it wasn't MMO though.
